Nerviano HER3-ADC is a preclinical antibody-drug conjugate (ADC) developed by Nerviano Medical Sciences. It consists of a monoclonal antibody targeting Human Epidermal Growth Factor Receptor 3 (HER3/ERBB3) conjugated to a proprietary duocarmycin-derivative payload. Unlike many current ADCs that utilize topoisomerase I inhibitors (e.g., deruxtecan), this candidate employs a DNA-alkylating duocarmycin payload to overcome resistance mechanisms, particularly in tumors that have progressed after treatment with Topo I-based ADCs. The duocarmycin derivative acts as a potent cytotoxic agent by alkylating DNA in the minor groove, leading to cell death in HER3-expressing cancer cells. It is currently being investigated for the treatment of therapy-resistant solid tumors, including breast cancer, colorectal cancer (CRC), and non-small cell lung cancer (NSCLC).
