Overview

NERx-329 is a first-in-class, small-molecule inhibitor of replication protein A (RPA) developed by NERx Biosciences. RPA is a critical single-stranded DNA (ssDNA) binding protein and a key sensor in the DNA damage response (DDR) pathway, particularly during replication stress. By competitively blocking the interaction between RPA and ssDNA, NERx-329 induces functional RPA depletion, replication fork degradation, and replication catastrophe, ultimately leading to apoptosis in cancer cells. NERx-329 is currently in IND-enabling development for the treatment of solid tumors, specifically lung cancer and ovarian cancer, both as a monotherapy and in combination with DNA-damaging agents like cisplatin or other DDR inhibitors.
