Overview

NERx-3392 is a first-in-class, small-molecule Ku-DNA binding inhibitor (Ku-DBi) being developed by NERx Biosciences for the treatment of various cancers, including esophageal cancer, head and neck cancer, and non-small cell lung cancer (NSCLC). It acts by blocking the interaction between the Ku70/Ku80 heterodimer and DNA, which prevents the recruitment and activation of the DNA-dependent protein kinase catalytic subunit (DNA-PKcs). This unique mechanism of action disrupts the non-homologous end joining (NHEJ) pathway of DNA double-strand break repair, thereby sensitizing cancer cells to DNA-damaging therapies such as ionizing radiation and chemotherapeutic agents like bleomycin.
