NES100 is an investigational intranasal drug product containing leucine-enkephalin, a naturally occurring endogenous peptide with analgesic properties. Developed using Molecular Envelope Technology (MET), NES100 is designed to enhance the delivery of enkephalin across the blood-brain barrier via a cartridge-based nasal device that propels the formulation along the olfactory nerve pathway. The MET nanotechnology protects and facilitates transport of this metabolically labile peptide directly to the brain, where it binds to delta opioid receptors to suppress pain. Preclinical studies have demonstrated analgesic potential without typical opioid-related adverse effects such as tolerance, withdrawal, respiratory depression, euphoria, or addiction. NES100 is being developed for acute and chronic non-cancer pain management as a non-addictive alternative to traditional opioids[1][4][5][6][7][8][9].
