Overview

The Nes2LR neoepitope vaccine is a preclinical peptide-based cancer immunotherapy designed to target renal cell carcinoma. It consists of a short, MHC-I restricted peptide neoepitope derived from a specific mutation in the Nesprin-2 protein (L4492R). When formulated with immunogenic particle-inducing adjuvants, such as cobalt-porphyrin-phospholipid (CoPoP) liposomes, the vaccine induces functional CD8+ T-cell responses. In murine renal adenocarcinoma (RENCA) models, it has demonstrated the ability to inhibit or eradicate tumors in subcutaneous, lung metastasis, and orthotopic settings, particularly when used in combination with immune checkpoint blockade therapy.
