Nestin shRNA is a short hairpin RNA-based therapeutic candidate designed to silence the expression of nestin, a class VI intermediate filament protein often used as a marker for neural stem cells and cancer stem cells. In malignant melanoma, nestin expression is associated with poor prognosis and advanced disease. By utilizing a silencing strategy, nestin shRNA reduces the levels of nestin protein, which in turn inhibits the AKT/MAPK signaling pathway, specifically decreasing the phosphorylation of AKT and p53. Preclinical studies have demonstrated that nestin knockdown leads to suppressed cell growth, reduced sphere formation, and inhibited invasion and metastasis in melanoma models.
