Nestin siRNA is a small interfering RNA therapeutic designed to target and silence the expression of nestin, a class VI intermediate filament protein. Nestin is a marker for progenitor/stem cells in the central nervous system and is highly expressed in various cancers, including pancreatic cancer, glioblastoma, and melanoma, where it correlates with tumor aggressiveness and metastasis. By silencing nestin mRNA, this siRNA inhibits cancer cell growth, migration, invasion, and sphere-forming ability. Preclinical studies in pancreatic cancer models have demonstrated that nestin siRNA, delivered via lipid nanoparticles (e.g., Invivofectamine), significantly reduces primary tumor growth and suppresses metastatic nodules in the liver and lungs, particularly when used in combination with gemcitabine.
