Overview

Nesuparib (JPI-547) is an orally administered small molecule that acts as a dual inhibitor of poly(ADP-ribose) polymerase (PARP) 1/2 and tankyrase 1/2. By inhibiting these enzymes—key members of the PARP family involved in DNA repair and the Wnt/β-catenin signaling pathway—nesuparib induces cell cycle arrest and apoptosis in tumor cells. It has demonstrated anti-tumor activity in preclinical models with BRCA-deficiency or homologous recombination repair deficiency (HRD), as well as Wnt-addicted cancers. Nesuparib is being developed primarily for ovarian cancer and pancreatic cancer but is also under investigation for other solid tumors including endometrial cancer. The drug was originally developed by Jeil Pharmaceutical and is currently being co-developed with Onconic Therapeutics[1][2][3][4][6].
