Nesvacumab is a fully human immunoglobulin G1 (IgG1) monoclonal antibody that specifically binds to and inhibits angiopoietin-2 (Ang2), a ligand for the TEK tyrosine kinase receptor (Tie2). By blocking Ang2, nesvacumab interferes with the Ang2-Tie2 interaction, thereby inhibiting angiogenesis and tumor cell proliferation. It was originally developed for cancer treatment but has also been investigated in ophthalmology for neovascular age-related macular degeneration (nAMD) and diabetic macular edema (DME). Clinical trials have shown that while nesvacumab can reduce central retinal thickness in DME, its combination with aflibercept did not provide additional visual acuity benefit over aflibercept monotherapy. Nesvacumab was developed by Regeneron Pharmaceuticals and Sanofi[1][3][4][5].
