Netazepide + rabeprazole is an oral combination drug consisting of **netazepide (YF476)**, a potent, highly selective, competitive, orally-active antagonist of the gastrin/CCK2 receptor, and **rabeprazole**, a proton pump inhibitor (PPI). Netazepide blocks the action of gastrin at the CCK2 (gastrin) receptor, thereby inhibiting gastrin-induced gastric acid secretion and the trophic (growth-promoting) effect of gastrin, especially on enterochromaffin-like (ECL) cells. Rabeprazole suppresses gastric acid secretion by irreversibly inhibiting the H+/K+-ATPase (proton pump) in the gastric parietal cells. The combination enhances suppression of gastric acid secretion more than either drug alone and may better counteract hypergastrinemia induced by PPIs by blocking gastrin's trophic effects. Indications explored have included gastric hypersecretion disorders, gastroesophageal reflux disease (GERD), and prevention of ECL cell hyperplasia.
