Netrin-1 neutralizing antibodies are a class of therapeutic agents designed to bind and inhibit netrin-1, a secreted protein that acts as a survival factor for cancer cells by preventing apoptosis mediated by dependence receptors such as DCC (Deleted in Colorectal Cancer) and UNC5 homologues. In many aggressive cancers, netrin-1 is overexpressed to evade programmed cell death. Neutralization of netrin-1 restores these apoptotic pathways, inhibits epithelial-mesenchymal transition (EMT), and modulates the tumor microenvironment by reducing angiogenesis and suppressing myeloid-derived suppressor cell (MDSC) infiltration. The most advanced clinical candidate in this class is NP137, a humanized IgG1 monoclonal antibody developed by NETRIS Pharma. NP137 has been evaluated in Phase 1 and Phase 2 clinical trials for advanced solid tumors, including endometrial and cervical cancers, and is also being explored for the treatment of chronic bone pain associated with arthritis and cancer.
