Overview

NEU-111 is a potent and highly selective oral allosteric inhibitor of tyrosine kinase 2 (TYK2), developed by Neuron23 for the treatment of immune-mediated inflammatory disorders. It acts by reducing signaling through several cytokine receptors, including those for interleukin (IL)-12, IL-23, and interferon (IFN)-alpha. Preclinical studies suggest that NEU-111 can achieve near-complete inhibition of TYK2 with low, once-daily dosing. TYK2 plays a central role in both innate and adaptive immune responses and is implicated in diseases such as inflammatory bowel disease (including ulcerative colitis and Crohn’s disease), systemic lupus erythematosus, rheumatoid arthritis, ankylosing spondylitis, and type 1 diabetes. As of early 2025, NEU-111 is being evaluated in Phase 1 clinical trials to assess its safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy volunteers[1][2][7].
