Drug intelligence / Profile preview
Neuroendocrine regulatory peptide (specifically NERP-4) is an endogenous 19-amino acid peptide derived from the granin protein VGF (secretogranin VII). It acts as a positive allosteric modulator of the sodium-coupled neutral amino acid transporter 2 (SNAT2/SLC38A2) on pancreatic beta cells. By enhancing the affinity of SNAT2, NERP-4 stimulates the uptake of neutral amino acids (such as glutamine, alanine, and proline) into beta cells, which in turn enhances mitochondrial ATP production, reduces oxidative and endoplasmic reticulum stress, and promotes glucose-stimulated insulin secretion (GSIS). Preclinical studies by the University of Miyazaki have shown that NERP-4 administration can restore impaired beta-cell maintenance and insulin secretion in diabetic mouse models, suggesting its potential therapeutic application in type 2 diabetes mellitus.
