Neurotrophin-3 plasmid DNA is a non-viral, naked plasmid DNA gene therapy encoding the neurotrophin-3 (NT-3) protein, developed by UNSW Sydney in collaboration with Cochlear Ltd. It is designed to promote the survival and targeted regrowth of cochlear (spiral ganglion) neurons toward the electrode array of a cochlear implant, thereby 'closing the neural gap' and improving the resolution and fidelity of auditory perception. The therapy is delivered locally into the cochlea during cochlear implant surgery using the BaDGE® (Bionic array Directed Gene Electrotransfer) platform, which utilizes close-field electroporation via the implant's electrode array to transfect mesenchymal cells lining the perilymphatic compartment. In clinical development, it is co-administered or co-expressed with a plasmid encoding Brain-Derived Neurotrophic Factor (BDNF). The combination was evaluated for safety and exploratory efficacy in the completed Phase I/II CINGT trial (ACTRN12618001556235) in adults with severe-to-profound sensorineural hearing loss undergoing cochlear implantation.
