Overview

A neutralizing antibody to hMCP-1 (human Monocyte Chemoattractant Protein-1, also known as CCL2) is a therapeutic modality designed to bind and neutralize the chemokine CCL2. CCL2 is a key regulator of monocyte/macrophage migration and infiltration into tissues during inflammatory processes and within the tumor microenvironment. By blocking the interaction between CCL2 and its primary receptor, CCR2, these antibodies aim to inhibit the recruitment of inflammatory monocytes and tumor-associated macrophages (TAMs). This pathway has been targeted for the treatment of various inflammatory diseases (such as rheumatoid arthritis and idiopathic pulmonary fibrosis) and oncology indications (such as metastatic castration-resistant prostate cancer and other solid tumors). Clinical candidates targeting this axis include carlumab (CNTO 888) and ABN912, though clinical development has faced challenges due to compensatory feedback loops that lead to a rebound increase in circulating CCL2 levels.
