New Path Molecular Research is developing a lead protein-drug conjugate (PDC) candidate that targets phosphatidylserine (PS), a phospholipid that is selectively exposed on the outer membrane of cancer cells and within the tumor microenvironment. Utilizing the company's "Beyond ADCs" platform, the drug is designed as an internalization-independent drug conjugate (IIDC). Unlike traditional antibody-drug conjugates (ADCs) that require endocytosis and lysosomal processing for payload release, this candidate is engineered to release its cytotoxic payload upon binding to PS in the extracellular space. This mechanism aims to bypass common resistance pathways associated with impaired cellular internalization. The lead program is currently focused on the treatment of colorectal cancer and pancreatic cancer, with potential for broader pan-tumor applications given that PS exposure is a common feature across various solid tumor types.
