Nexatecan is a phenolic camptothecin-based small molecule cytotoxic agent developed by Intocell as a specialized payload platform for antibody-drug conjugates (ADCs). It utilizes Intocell's proprietary OHPAS (Ortho-Hydroxy Phenyl Acetate Spacer) technology, which is designed to optimize the stability and release kinetics of the drug. As a camptothecin derivative, nexatecan functions as a topoisomerase I inhibitor, inducing DNA damage and apoptosis in cancer cells. The platform is engineered to provide superior efficacy and a broader therapeutic window compared to existing ADC payloads like deruxtecan, specifically aiming to reduce systemic toxicity and common side effects such as interstitial lung disease (ILD).
