Nexturastat A is a potent and highly selective small molecule inhibitor of histone deacetylase 6 (HDAC6). It was developed to improve upon the narrow therapeutic window of pan-HDAC inhibitors by specifically targeting the HDAC6 isoform, which is involved in various non-epigenetic cellular processes and immune regulation. Research indicates that Nexturastat A can modulate the tumor microenvironment by down-regulating the expression of immune checkpoint proteins such as PD-L1 and PD-L2, reducing the presence of immunosuppressive M2 macrophages, and increasing T-cell infiltration and memory. It is primarily being investigated in preclinical models as a priming agent to sensitize immunologically "cold" tumors to immune checkpoint blockade therapies, such as anti-PD-1 and anti-CTLA4, particularly in melanoma.
