Overview

Nezastomig + cemiplimab is an investigational combination immunotherapy being developed by Regeneron Pharmaceuticals and Bayer. The combination consists of nezastomig (REGN5678), a first-in-class bispecific antibody that targets prostate-specific membrane antigen (PSMA) and CD28, and cemiplimab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1). Nezastomig provides a co-stimulatory signal to T-cells by bridging PSMA-expressing tumor cells and CD28 on T-cells, while cemiplimab blocks the PD-1/PD-L1 inhibitory pathway. This dual approach is designed to enhance anti-tumor T-cell activation and proliferation, particularly in patients with metastatic castration-resistant prostate cancer (mCRPC) and clear cell renal cell carcinoma (ccRCC).
