NFκB-siRNA (nuclear factor kappa B small interfering RNA) refers to a class of short interfering RNA therapeutics designed to target and silence the expression of the NF-κB transcription factor or its specific subunits (such as p65/RelA or p50/NFKB1). NF-κB is a key transcription factor that regulates genes involved in cell survival, proliferation, inflammation, and immune responses, and its constitutive activation is associated with tumor progression, chemoresistance, and inflammatory diseases. To overcome the challenges of naked siRNA delivery, such as rapid degradation and poor cellular uptake, researchers have developed various nanocarriers, including gelatin nanocarriers (NS-GNCs) developed by the University of Hawaii, gold nanoparticles, and reducible branched polyethylenimine (rBPEI) nanoparticles, to deliver NFκB-siRNA effectively to target tissues like tumors or corneal neovascularization sites.
