NF-56-EJ40 is a potent and selective small molecule antagonist of the succinate receptor 1 (SUCNR1), also known as GPR91. Succinate, a key mitochondrial metabolite, often accumulates in the hypoxic and nutrient-deprived tumor microenvironment (TME) of solid tumors, where it acts as a signaling molecule to promote an immunosuppressive landscape. NF-56-EJ40 has been demonstrated to block succinate-induced proliferation and function of regulatory T cells (Tregs), which are central components of the TME that obstruct anti-tumor immunity. By inhibiting the SUCNR1 signaling pathway, NF-56-EJ40 has the potential to reduce Treg infiltration and suppressive activity, thereby enhancing the effectiveness of T cell immunotherapies and restoring anti-tumor immune responses. It is primarily utilized as a research tool to investigate the role of the succinate-SUCNR1 axis in oncology and inflammatory diseases.
