NF-kB knockout skin-derived mesenchymal stem cells are a genetically engineered form of skin-derived mesenchymal stem cells (S-MSCs) in which the NF-kappaB signaling pathway has been permanently disrupted, typically through gene editing or knockout of critical NF-kB subunits (such as IKK or p65/RelA). This modification is designed to remove or sharply reduce the activity of the NF-kB family of transcription factors, which play key roles in controlling inflammatory and immune responses, cell survival, differentiation, and migration. The primary rationale for developing these cells is to alter S-MSC behavior for specific therapeutic purposes, typically by reducing pro-inflammatory signaling and potentially enhancing differentiation into desired cell types (e.g., osteoblasts, chondrocytes, myocytes) for regenerative medicine or to lower inflammatory side effects in cell therapy. Skin-derived MSCs themselves are a promising cell source for therapy due to their immunomodulatory properties and ease of expansion. Targeted NF-kB knockout is expected to modify their cytokine profile, reduce their pro-inflammatory potential, and change their differentiation and migratory behavior. Currently, most NF-kB silencing or knockout work in MSCs is preclinical, and specific commercial or clinical-stage products under this exact name are not described in the literature as of 2025[2][5][4][1].
