NF2 shRNA is a research-grade short hairpin RNA (shRNA) construct designed to silence the expression of the neurofibromin 2 (NF2) gene, which encodes the tumor suppressor protein Merlin. In oncological research, particularly concerning malignant mesothelioma, NF2 shRNA is utilized to model the initiation of tumors by disrupting the Hippo signaling pathway. The knockdown of NF2 leads to the dephosphorylation and subsequent nuclear translocation of Yes-associated protein (YAP), which drives the transcription of genes involved in cell proliferation and transformation. This tool is primarily employed in laboratory settings to study the molecular mechanisms of mesothelial cell transformation and to identify downstream effectors, such as PLCB4, that may serve as therapeutic targets.
