NFlection Therapeutics is developing a proprietary 'soft' (metabolically labile) topical MEK inhibitor for the treatment of keratinocytic epidermal nevi and nevi sebacei, which are congenital birthmarks. This small molecule is designed to inhibit the Ras/Raf/MEK/ERK pathway locally within the skin lesions while rapidly degrading upon reaching systemic circulation to minimize potential side effects. Preclinical studies have demonstrated dose-dependent suppression of pERK, a biomarker of pathway activation, in human nevus sebaceous and epidermal nevus explants. The drug is specifically engineered to provide high local efficacy while avoiding the systemic toxicities typically associated with MEK inhibition.
