nfP2X7 refers to a non-functional variant of the P2X purinoceptor 7 (P2X7) receptor. Unlike the wild-type P2X7, which can form an apoptotic pore upon prolonged ATP stimulation and induce cell death, the nfP2X7 variant exposes a normally hidden epitope and is unable to initiate apoptosis but retains channel activity. This variant is robustly expressed on many types of cancer cells but not on normal healthy tissues. Targeting nfP2X7 has emerged as a promising therapeutic strategy in oncology due to its tumor-specific expression and role in cancer cell survival. Therapeutic approaches include monoclonal antibodies (such as BIL010t for basal cell carcinoma), CAR-T cell therapies targeting ovarian and other solid cancers, and dendritic-cell based immunotherapies. These strategies exploit the selective expression of nfP2X7 on tumor cells to induce anti-tumor immune responses or direct cytotoxicity while sparing normal tissues[3][4][5][8].
