NFYC-AS1 gapmer refers to a series of research-grade antisense oligonucleotides (ASOs), specifically the GAP1 through GAP6 sequences sourced from QIAGEN, designed to target and silence the long non-coding RNA (lncRNA) NFYC-AS1. The mechanism of action involves RNase-H-mediated cleavage of the complementary mature RNA transcript upon binding. NFYC-AS1 is a known molecular vulnerability in various malignancies, including acute myeloid leukemia (AML), where it is overexpressed and supports cell cycle progression and survival. Research indicates that depletion of NFYC-AS1 using these gapmers can alter chromatin conformation at the chr19p13.3 locus and decrease the expression of essential genes involved in myeloid differentiation, leading to growth inhibition in leukemic blasts. These gapmers are primarily used as research tools to study lncRNA function and therapeutic potential in cancer biology.
