Overview

NG-347 is a **systemically delivered oncolytic virus-based gene therapy** from PsiOxus Therapeutics for the treatment of solid tumors. Built on the company's T-SIGn platform, it is designed to selectively infect tumors after intravenous administration and convert tumor cells into localized producers of immunostimulatory payloads. NG-347 is armed with three transgenes: secreted interferon alfa, secreted macrophage inflammatory protein 1 alpha, and membrane-bound CD80. Its intended mechanism is to generate intratumoral interferon alfa signaling independent of defective STING-pathway activation, recruit inflammatory immune cells through CCL3 activity, and enhance T-cell costimulation through CD80, thereby promoting antitumor immune responses in solid tumors. Publicly described development places the program in preclinical development with a planned clinical start around 2019.
