NG-348 is an investigational armed oncolytic virus therapy developed for the treatment of solid tumors. It is a transgene-modified variant of enadenotucirev, a chimeric adenovirus engineered to selectively infect and replicate within tumor cells. Using PsiOxus Therapeutics' proprietary Tumor-Specific Immuno-Gene Therapy (T-SIGn) platform, NG-348 encodes two immunomodulatory membrane-integrated T-cell-engaging (MiTe) proteins. When expressed together on the surface of infected tumor cells, these proteins provide both T-cell receptor activation (signal 1) and costimulatory activation (signal 2), enabling polyclonal activation of tumor-infiltrating CD4 and CD8 T-cells in an antigen-independent manner. This local immune stimulation aims to drive potent anti-tumor responses while minimizing off-target effects in healthy tissues[1][2][6]. The drug was originally developed by PsiOxus Therapeutics (now Akamis Bio) and licensed to Bristol Myers Squibb for further clinical development[1][2]. As of the latest updates, it has been evaluated in phase I trials for solid tumors[2].
