Overview

A combination investigational regimen that includes: - **NG-350A**: a next-generation adenoviral vector engineered to express an agonist anti-CD40 monoclonal antibody, selectively replicating within tumors. NG-350A drives intratumoral expression of CD40 agonist antibody, activating antigen-presenting cells (APCs) to recruit T-cells for enhanced anti-tumor immune response[1][3][5]. - **Ipilimumab**: a monoclonal antibody targeting Cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), inhibiting immune checkpoint signaling and boosting T-cell mediated anti-tumor activity. - **Nab-paclitaxel**: a nanoparticle albumin-bound formulation of paclitaxel, disrupting microtubule dynamics in cancer cells, leading to apoptosis. This formulation enhances drug delivery by exploiting albumin-mediated transport[4][6][8]. - **Gemcitabine**: a nucleoside analog used as a chemotherapy agent, inhibiting DNA synthesis in dividing tumor cells[4][6][8]. The combination is being studied for **metastatic pancreatic adenocarcinoma** in clinical trials, with NG-350A providing immunomodulation through intratumoral CD40 activation, ipilimumab acting as a checkpoint inhibitor, and nab-paclitaxel plus gemcitabine serving as cytotoxic agents[1]. NG-350A is developed by Akamis Bio and investigated in collaboration with the Parker Institute for Cancer Immunotherapy and the Cancer Research Institute[1][3]. The combination is not yet approved and is studied in the Phase 1b REVOLUTION trial.
