Overview

NG-641 + nivolumab is an investigational combination therapy being studied for the treatment of advanced or metastatic epithelial tumors. NG-641 is a next-generation, blood-stable, transgene–armed T-SIGn adenoviral vector developed to selectively infect and replicate within cancer cells. It expresses four immunostimulatory transgenes that crosslink T cells to fibroblast activating-protein–positive cancer-associated fibroblasts, thereby activating the immune system. The vector also induces T-cell migration via C-X-C motif chemokine ligand 9 (CXCL9) and C-X-C motif chemokine ligand 10 (CXCL10) expression and enhances T-cell activation through interferon-alpha2, promoting tumor cell killing by antigen-specific T cells[2][8]. Nivolumab (Opdivo) is a programmed cell death protein 1 (PD‑1) blocking monoclonal antibody that restores anti-tumor immunity by inhibiting PD‑1-mediated immune suppression[4]. The combination is under evaluation in the phase 1a/b NEBULA trial for patients with previously treated metastatic or advanced epithelial tumors who have progressed after at least one line of systemic therapy[5][3].
