Overview

NG-796A is a selective, small molecule antagonist of the arginine vasopressin receptor 1A (AVPR1A), originally developed by Neurogen Corporation. The vasopressin system, particularly the V1a receptor subtype, is a key regulator of social behavior, stress responses, and emotional processing in the central nervous system. NG-796A was designed to cross the blood-brain barrier and block the effects of vasopressin at the V1a receptor, with the primary therapeutic goal of treating social anxiety disorder (SAD) and generalized anxiety disorder (GAD). By inhibiting V1a-mediated signaling, the drug aimed to reduce the physiological and psychological symptoms associated with social distress and chronic anxiety. Clinical development reached Phase 2 trials; however, following the acquisition of Neurogen by Ligand Pharmaceuticals in 2009, the development of NG-796A was not prioritized and the program was subsequently terminated.
