Overview

NG-nitro-l-arginine methyl ester (L-NAME) is a cell-permeable, more soluble analog of arginine that acts as a competitive and slowly reversible inhibitor of nitric oxide synthase (NOS), particularly endothelial NOS (eNOS). By inhibiting NOS, L-NAME reduces the synthesis of nitric oxide (NO), a key signaling molecule involved in vasodilation and vascular homeostasis. This compound has been widely used in experimental research to study the physiological and pathological roles of NO. In animal models, chronic administration induces hypertension by suppressing NO-mediated vasodilation and can be used to model cardiovascular diseases such as hypertension and cardiac remodeling[1][2][3][6]. L-NAME has also been investigated for its effects on capillary leak syndrome during interleukin 2 therapy in cancer models[2], as well as for its ability to prevent glutamate toxicity via inhibition of neuronal NOS[5]. It is not approved for clinical use but remains an important pharmacological tool in preclinical studies.
