NG.IL-24 (also known as Enhanced Therapeutic Cytokine or Superkine) is a next-generation, genetically engineered form of interleukin-24 (IL-24/mda-7) developed by InterLeukin Combinatorial Therapies. This designer cytokine was engineered to improve upon wild-type IL-24 through two primary modifications: the replacement of the endogenous secretory motif with an insulin secretory motif to enhance protein secretion, and a K122R amino acid substitution to increase protein stability. The resulting molecule, often referred to as M7S, maintains the cancer-specific activity of IL-24, inducing programmed and immunogenic cell death, inhibiting angiogenesis, and sensitizing tumor cells to conventional therapies. It is being investigated for the treatment of various cancers, including glioblastoma, melanoma, and carcinomas of the breast, lung, and pancreas, and can be delivered as a purified protein, via viral vectors, or through engineered T cells.
