Overview

NG12-1 is a topically applied adrenergic vasoconstrictor formulation developed to prevent radiodermatitis in cancer patients undergoing radiotherapy. Developed by Suzhou NG Biomedicine and Vaso Dynamics, the drug utilizes an aqueous alcohol-based vehicle to deliver high concentrations of an adrenergic agonist through the stratum corneum to the subcutaneous vasculature. By inducing transient local skin hypoxia during radiation sessions, NG12-1 reduces the formation of reactive oxygen species (ROS) and inhibits the fixation of ROS-induced DNA damage in epidermal stem cells, thereby protecting the skin from radiation-induced toxicity. Clinical studies, including a Phase IIa trial in breast cancer patients (NCT01263366), have demonstrated significant reductions in the severity of radiodermatitis without systemic adverse effects such as blood pressure elevation or tachycardia.
