NG2 CAR-T cells + DLL3 CAR-T cells is a combination cellular immunotherapy approach involving the co-administration of two distinct chimeric antigen receptor (CAR) T-cell products. One product targets Chondroitin Sulfate Proteoglycan 4 (CSPG4), also known as NG2, a cell surface antigen frequently overexpressed in various solid tumors including melanoma, glioblastoma, and certain neuroendocrine cancers. The second product targets Delta-like ligand 3 (DLL3), an inhibitory Notch ligand that is highly expressed on the surface of small cell lung cancer (SCLC) and other neuroendocrine tumor cells but has minimal expression in normal tissues. By combining these two CAR-T cell populations, the therapy aims to overcome tumor heterogeneity and reduce the risk of antigen escape, a common mechanism of resistance in single-target CAR-T therapies. This dual-targeting strategy is primarily being investigated for the treatment of aggressive neuroendocrine tumors where both targets may be present.
