NG2xUCHT-1 is a bispecific antibody (bsAb) designed to induce T-cell-mediated immunity against melanoma. It targets the neuron-glial antigen 2 (NG2), also known as melanoma-associated chondroitin sulfate proteoglycan (MCSP) or CSPG4, which is selectively overexpressed in approximately 90% of melanoma cases. The antibody utilizes the UCHT-1 clone as a high-affinity CD3 binder to recruit and activate T cells directly at the tumor site, leading to target-dependent tumor cell lysis. Developed by researchers at the University Hospital Tübingen, NG2xUCHT-1 serves as a high-affinity benchmark in the development of T-cell engagers, alongside variants like NG2xM18 which aim to optimize the balance between antitumor efficacy and cytokine release syndrome (CRS).
