Overview

NGB 2904 is a potent and highly selective small molecule antagonist of the dopamine D3 receptor, with a Ki of 1.4 nM for D3 and >200-fold selectivity over D2 and other monoaminergic receptors[1][7]. It has been widely used as a research tool in animal models to study addiction and motivational behaviors. Systemic administration inhibits cocaine self-administration under progressive-ratio reinforcement schedules, suppresses cocaine- and cue-induced reinstatement of drug-seeking behavior, and attenuates cocaine-induced brain stimulation reward responses, without affecting non-drug reward behaviors[2][4][6][8]. The compound was originally developed by Neurogen Corp. and is used primarily in laboratory research settings for elucidating dopaminergic mechanisms in addiction[9].
