NGC-Iri is a nanogel-encapsulated formulation of the cytotoxic agent irinotecan, developed by NanoGen Biopharmaceutical. Irinotecan is a camptothecin derivative and a prodrug that is converted by carboxylesterases to its active metabolite, SN-38. SN-38 acts as a potent inhibitor of DNA topoisomerase I, an enzyme essential for DNA replication and transcription. By stabilizing the cleavable complex between topoisomerase I and DNA, SN-38 prevents the religation of DNA strands, leading to double-strand breaks and subsequent apoptosis in rapidly dividing tumor cells. The NGC (nanogel) delivery system is designed to improve the pharmacokinetic profile of irinotecan by enhancing its solubility and stability, potentially increasing tumor accumulation via the enhanced permeability and retention (EPR) effect while reducing systemic toxicity compared to conventional irinotecan formulations.
