Overview

NGcGM3 (N-glycolyl-GM3) is a N-glycolylated ganglioside, a type of glycosphingolipid, that functions as a tumor-associated antigen. It is uniquely expressed on various cancer cells, including ovarian, breast, melanoma, lymphoma, bladder, neuroblastoma, retinoblastoma, Wilms' tumor, brainstem glioma, and lung cancers, but is absent from healthy human tissues due to a genetic deletion in humans. This makes it a neoantigen, often resulting from metabolic incorporation from dietary sources. Its specific expression pattern makes it a promising target for immunotherapeutic strategies such as CAR T cell therapies, therapeutic vaccines (e.g., NGcGM3/VSSP), and monoclonal antibodies. NGcGM3 also acts as an endogenous ligand for CD1d and can modulate immune cell functions, including down-modulating CD4 expression on T lymphocytes and influencing dendritic cell differentiation.
