Overview

NGI-1 (also known as ML414) is a small molecule research compound that functions as an inhibitor of the oligosaccharyltransferase (OST) complex. It specifically targets the catalytic subunits STT3A and STT3B, which are responsible for the N-glycosylation of proteins. By inhibiting this process, NGI-1 disrupts the maturation and stability of various glycoproteins, including viral proteins and receptor tyrosine kinases such as EGFR. It has demonstrated broad-spectrum antiviral activity against flaviviruses, including Dengue, Zika, West Nile, and Yellow Fever viruses, by blocking viral RNA replication. In oncology, NGI-1 has shown potential in treating non-small-cell lung cancer (NSCLC) by overcoming resistance to EGFR inhibitors, enhancing radiosensitivity in glioma, and inducing apoptosis in multiple myeloma cells. NGI-1 is currently used exclusively as a research tool and is not approved for human use or under active commercial development.
