Overview

The NGL-1 targeting peptide is a research-stage, functionally antagonistic small peptide designed to inhibit Netrin G1 Ligand (NGL-1, also known as LRRC4C). NGL-1 is a synaptic protein that is ectopically expressed in the tumor microenvironment (TME) of pancreatic ductal adenocarcinoma (PDAC), specifically on cancer-associated fibroblasts (CAFs) and immune cells such as myeloid and T cells. By targeting NGL-1, the peptide reduces the production of immunosuppressive cytokines and restores anti-tumor immune activity, effectively modulating the TME in a TGF-β-dependent manner. Research conducted at institutions like Fox Chase Cancer Center suggests that NGL-1 inhibition can phenocopy the effects of chemotherapy and improve survival in preclinical models of pancreatic cancer.
