Overview

NGM831 is a human monoclonal antibody developed as an antagonist of ILT3 (also known as LILRB4, leukocyte immunoglobulin-like receptor subfamily B member 4). It is designed to block the interaction between ILT3 and its ligands, including fibronectin, a key component of the tumor stroma. By inhibiting this pathway, NGM831 aims to reprogram myeloid cells from an immune-suppressive state to a stimulatory phenotype within the tumor microenvironment. This mechanism is intended to enhance anti-tumor immunity by promoting antigen presentation and T cell activation. Preclinical studies have shown that NGM831 can synergize with pembrolizumab (an anti-PD-1 checkpoint inhibitor) to further stimulate immune responses against tumors. The drug is being investigated for use in advanced or metastatic solid tumors across multiple cancer types[3][5][6][7].
