Overview

NGM936 is a bispecific T-cell engager (TCE) developed by NGM Biopharmaceuticals for the treatment of hematologic malignancies, including acute myeloid leukemia (AML) and multiple myeloma. The molecule is a monovalent scFv-Fab bispecific antibody constructed on an effectorless human IgG1 (LALA) backbone with a knob-in-hole Fc design to ensure correct heterodimeric assembly. NGM936 targets Immunoglobulin-like transcript 3 (ILT3, also known as LILRB4), an inhibitory receptor that is highly expressed in monocytic AML and multiple myeloma but absent on normal hematopoietic stem cells and non-monocytic blood cells. By simultaneously binding ILT3 on tumor cells and CD3 on T cells, NGM936 facilitates T-cell dependent cytotoxicity (TDCC) against malignant cells. Preclinical data indicate potent anti-tumor activity with minimal cytokine release. As of early 2024, NGM936 is in preclinical development, and the company is seeking partners for its clinical advancement.
