Overview

NgR(310)ecto-Fc is a recombinant fusion protein designed to promote axonal regeneration and neuroprotection in the central nervous system. It consists of the ligand-binding ectodomain of the Nogo-66 receptor (NgR1, specifically amino acids 27-310) fused to the Fc constant region of an immunoglobulin (typically rat IgG1 in preclinical studies). The molecule functions as a decoy receptor that binds to and neutralizes three major myelin-associated inhibitors (MAIs) of axonal growth: Nogo-66 (a domain of Reticulon-4), myelin-associated glycoprotein (MAG), and oligodendrocyte myelin glycoprotein (OMgp). By sequestering these inhibitors, NgR(310)ecto-Fc prevents them from interacting with endogenous axonal NgR1, thereby blocking the signaling pathways that lead to growth cone collapse and inhibition of neurite outgrowth. Developed primarily at the Strittmatter Laboratory at Yale University, it has demonstrated neuroprotective effects and enhanced axonal sprouting in preclinical models of spinal cord injury, stroke, glaucoma, multiple sclerosis, and Alzheimer's disease.
