NGR-hTNF + cisplatin is a combination regimen composed of **NGR-hTNF**, a recombinant fusion protein consisting of human tumor necrosis factor-α (TNF) fused to the tumor-homing peptide asparagine–glycine–arginine (NGR), and **cisplatin**, a platinum-based chemotherapeutic agent. The NGR motif targets aminopeptidase N (CD13) isoforms overexpressed on angiogenic tumor endothelium, enhancing selective delivery of TNF to tumor vasculature and rapidly increasing vascular permeability. This facilitates the enhanced uptake of cisplatin (and other chemotherapeutics) into tumors. NGR-hTNF promotes apoptosis of tumor endothelial cells, reduces interstitial fluid pressure, enables improved chemotherapeutic delivery, and may promote antitumor immune responses. In clinical studies, the combination shows promising antitumor activity and favorable tolerability in patients with advanced chemoresistant malignancies[1][2][4].
