Overview

NGR-hTNF + doxorubicin is an investigational combination therapy consisting of **NGR-hTNF**, a recombinant fusion protein of the tumour-homing peptide (asparagine–glycine–arginine) conjugated to **human tumor necrosis factor-α (TNF-α)**, and **doxorubicin**, an anthracycline chemotherapy. **NGR-hTNF** selectively targets the tumor vasculature by binding to aminopeptidase N (CD13), which is overexpressed on tumor blood vessels, resulting in vascular damage that enhances both immune cell infiltration and intratumoral accumulation of doxorubicin. The combination is designed to leverage this effect, aiming to improve the efficacy of doxorubicin against refractory or resistant solid tumors and certain relapsed cancers. Clinical studies have demonstrated manageable toxicity, no significant drug–drug pharmacokinetic interaction, and promising disease control and response rates in heavily pre-treated populations, particularly in advanced ovarian cancer and relapsed small cell lung cancer[1][2][3].
