Overview

This drug is a **combination therapy** consisting of NGR-hTNF, oxaliplatin, and capecitabine, evaluated in oncology clinical research, primarily for the treatment of advanced or metastatic solid tumors. NGR-hTNF is a recombinant cytokine, in which human TNF-α is fused to the CNGRC peptide motif, enabling the **selective targeting of angiogenic tumor vasculature** via binding to a CD13 isoform overexpressed in tumor blood vessels. This targeting enhances vascular permeability, improves delivery of chemotherapeutic agents, impairs tumor endothelial barrier function, induces **endothelial and tumor cell apoptosis**, and promotes an immune-supportive microenvironment. Oxaliplatin is a third-generation platinum-based chemotherapeutic agent; it acts by **crosslinking DNA**, leading to cell death, and is mainly used in colorectal, gastric, and other gastrointestinal cancers. Capecitabine is an oral prodrug converted to 5-fluorouracil (5-FU) in vivo, which **inhibits thymidylate synthase**, leading to impaired DNA synthesis and apoptosis; it is commonly used for colorectal and breast cancer. This triple combination is being studied particularly for conditions such as advanced colorectal cancer and other solid tumors, leveraging NGR-hTNF’s ability to increase drug delivery and synergize with oxaliplatin and capecitabine for antitumor effects[1][3][4][2][5].
