NGR-TNF (also known as NGR-hTNF) is a vascular targeting recombinant fusion protein consisting of human tumor necrosis factor-alpha (TNF-alpha) fused to the tumor-homing peptide CNGRC. This peptide selectively binds to aminopeptidase N (CD13), which is highly expressed on the endothelial cells of tumor neovasculature. By concentrating TNF-alpha at the tumor site, NGR-TNF induces changes in vascular permeability at low doses and direct vascular damage at higher doses, thereby improving the delivery of chemotherapy or exerting direct antitumor effects while minimizing systemic TNF toxicity. The European Organisation for Research and Treatment of Cancer (EORTC) conducted the Phase I EORTC 16041 trial to evaluate its safety and optimal dosing in patients with advanced solid tumors.
