Overview

NH104 is a small molecule dual orexin receptor antagonist (DORA) developed by Jiangsu Nhwa Pharmaceutical. It is primarily being investigated for the treatment of insomnia and depression. By antagonizing both orexin 1 (OX1) and orexin 2 (OX2) receptors, NH104 inhibits the wake-promoting effects of the orexin system, thereby facilitating sleep onset and maintenance. This mechanism of action is distinct from traditional sedative-hypnotics like benzodiazepines, potentially offering a better safety profile regarding next-day grogginess and dependency. As of early 2024, the candidate is in Phase 1 clinical development for insomnia.
