Overview

NHI-2 is a **small molecule inhibitor** of the enzyme **lactate dehydrogenase A (LDHA)**, designed to directly target cancer cell metabolism. It reduces lactate production in various cancer cell lines, including HeLa, LPC006, and PANC-1, by inhibiting LDHA activity. Preclinical studies have shown that NHI-2 disrupts glycolysis, decreases cellular ATP and NADH/NAD+ ratio, and inhibits cancer cell growth. It is particularly cytotoxic against pancreatic cancer cell lines and has been found to enhance the antitumor activity of gemcitabine in vitro. NHI-2 may also inhibit ferroptosis. The compound is under investigation as an antineoplastic agent but is not yet in clinical development[3][1].
